Thaw Sperm in Straws

Reagents, supplies, and equipment:

• Paper clip for plunger
• IVF dish
• Scissors
• 100% ethanol
• Room temperature water bath

Thawing Sperm in Straws:

1. Remove straw from LN2 storage.
2. Immerse straw in 37 degree temperature water for ten minutes.
3. Wipe off straw with ethanol.
4. Holding the straw firmly, cut off the seal, leaving about half of the cotton plug in place.
5. Using a metal rod as a plunger, push against the remaining portion of the cotton plug and expel the sperm fraction CARD drop in IVF dish.
